Two-Dimensional Polyacrylamide Gel Electrophoresis – A Practical Perspective
نویسندگان
چکیده
Twodimensional electrophoresis, abbreviated as 2-DE, is one of the most powerful and common tools for separation and fractionation of complex protein mixture extracted from tissues, cells, and other biological specimens. It is an orthogonal technique that allows separation of thousands of proteins in one gel and in a two tandem electrophoretic steps where a major proportion of proteins can be resolved for further analysis. Since the pioneer development and modification of high resolution 2-DE by (O'Farrell, 1975), the usage of this technique is increasing and proven to be effective. In fact, the modification set by O’Farrell empowered this technique to resolve up to 5000 spots in a uniform distribution and on a single gel. Protein spot can be seen as little as one disintegration per min by autoradiology or recent quantitative dye. This high reproducibility of separation allows comparative matching for detection of dysregulated proteins.

A Simpler and Faster Version of Two-Dimensional Gel Electrophoresis Using Vertical, Mini SDS-PAGE Apparatus
We have modified one of the most useful methods of protein separation; namely, two dimensional gel electrophoresis (2-DE). This modified version of 2-DE is not only simpler and easier but also faster than all the currently available methods. In this method, isoelectric focusing is carried out in the first dimension using a vertical sodium dodecyl sulfate polyacrylamide gel electrop...
متن کاملTwo–Dimensional Gel Electrophoresis of Ceolomic Fluid of Eisenia foetida Earthworm
Earthworms possess antioxidant, antibacterial, antitumor, and hemolytic properties. To recognize the molecules responsible for various biological activities of earthworm’s coelomic fluid, a detailed knowledge about its protein contents is required. The aim of this study was to characterize the proteins present within the coelomic fluid of Eisenia foetida earthworm. Polyacrylamide-gel-elec...

متن کاملDouble-label reductive methylation of tissue proteins for precision two-dimensional polyacrylamide-gel electrophoretic analysis.
Reductive methylation has been used to radioactively label crude-extract proteins with 3H or 14C. The procedure achieved good isotope incorporation and resolution of proteins on two-dimensional polyacrylamide gels. It allows high-precision comparison of tissue samples by double-labelling and should facilitate the study of tissue proteins by two-dimensional gel electrophoresis.

Two-dimensional gel electrophoresis (2-DE) methods such as two-dimensional polyacrylamide gel electrophoresis (2D-PAGE; O’Farrell (1975)) and two-dimensional difference gel electrophoresis (2D-DIGE; Ünlü et al. (1997)) are popular techniques for protein separation because they allow researchers to characterize quantitative protein changes on a large scale. Thus, 2-DE is frequently used as an in...
